Our laboratory focuses on mechanistic discovery to understand the metabolic and molecular mechanisms of androgen synthesis and androgen receptor gain-of-function that lead to resistance to hormonal therapy.  Specific areas include: 1) Metabolic and genetic changes required for hormone therapy resistance in prostate cancer and tumor progression 2) Clinical validation in patients and clinical trials using innovative approaches 3) Animal models of advanced prostate cancer for translational and therapeutic studies 4) Identifying targets for the development of new pharmacologic therapies We discovered the first example of a gain-of-function in a steroid-synthesizing enzyme that enables prostate cancer resistance to hormonal therapy (Chang, et al. Cell. 2013;154:1074-84). We also recently discovered that abiraterone works by conversion to a more active steroidal metabolite (Li, et al. Nature. 2015;523:347-51), that metabolism is pharmacologically modifiable to optimize therapy (Li, et al. Nature. 2016;533:547-51) and that these events are a class effect of steroidal androgen synthesis inhibitors (Alyamani, et al. Cell Chem Biol.2017;24:825-32) and genetic determination of metabolite generation (Alyamani, et al. J Clin Invest. 2018;128:3333-40). This position is ideal for an individual with a strong interest in rapid translation of basic mechanistic discoveries to the bedside as this is a principal goal of the Sharifi Laboratory. For example, we have shown that our discovery of a gain-of-function in a steroid-synthesizing enzyme is a predictive biomarker of poor outcomes after hormonal therapy (Hearn, et al. Lancet Oncol. 2016;17:1435-44; JAMA Oncol. 2018;4:558-62).   We are currently evaluating this biomarker in an active clinical trial and are pursuing similar mechanisms and developing new treatment modalities based on these discoveries. The position will provide a unique and multidisciplinary exposure to tumor metabolism, molecular oncology, drug development and clinical trials. The ideal candidate has a Ph.D. or a visiting scholar in biochemistry, chemistry or molecular biology; has the appropriate expertise in discovery of molecular, biochemical or chemical mechanisms; and is highly driven.  Outstanding verbal and communication skills are required. The general aim of our lab is to delineate how the tumor and stroma/immune cell populations adapt and interact during the development of drug resistance. We will be utilizing novel single cell analytics including single cell omics, novel multiplex immunoflu orescence, and high complexity co-culture models to model the immune vs. tumor interaction in patient tumors. We work closely with multiple collaborators to use in vivo models of cancer and patient-derived tumor xenografts to validate our hypotheses. Enthusiastic candidates with a recent PhD or a visiting scholar  with strong expertise in molecular and cellular biology are encouraged to apply. The ideal candidate should have: 1) first author publication in peer-reviewed journal, 2) extensive experience in wet lab molecular and cell biology work (experience in organoid or other co-culture models will be a strong plus), 3) working knowledge in cancer biology and/or immunobiology and 4) strong written and oral English communication skills. Our laboratory is investigating novel therapeutics for patients with leukemia and solid tumors.  A focus of our laboratory is to investiga te the function of onco-embryonic antigens, such as ROR1, which are selectively expressed by tumor cells and that contribute to malignant behavior. This also involves development and testing of novel agents that can target such antigens and/or their associated signaling pathways that contribute to the survival, growth, metastatic potential, and/or cancer stemness of neoplastic cells, even in the setting of conventional anti-cancer therapy. These studies can result in the development of more-effective therapies for patients with cancer. Applicants must have a PhD or visiting scholar, strong analytical skills, proficiency in oral and written scientific presentations, and a demonstrated commitment to a career in cancer research. Postdoctoral position is preferably offered to candidates who have recently received their PhD or have three years or less of post graduate education. Such questions have important implications for understanding and treating a wide range of human diseases including autoimmunity, infection and cancer. Currently, we are looking for visiting scholars and postdoctoral fellows to work in two program areas. The first focuses on how epigenetic mechanisms regulate chromatin structure in the context of B cell development. The second project is focused on understanding the genetic and epigenetic regulatory mechanisms of germinal center responses. Both project areas are well-established and supported by a highly interactive group of scientists and extensive core resources. Job qualifications: we are looking for graduate students in immunology and/or molecular biology. We will also consider graduates of bioinformatic or genetic programs interested in complementing their computational skills with bench experimentation. We are seeking a dynamic individual who will be responsible, as a member of a team, for designing and executing studies focused on understanding and targeting altered RNA processing mechanisms in pancreatic cancer. As the most common form of pancreatic cancer, pancreatic ductal adenocarcinoma (PDAC) has emerged as the 3rd leading cause of cancer death in the United States, in part because this malignancy is highly resistant to chemotherapeutic intervention. Recently, it’s been shown that PDAC is composed of distinct molecular subtypes, based on RNA expression and patient outcome, underscoring that much remains to be understood about RNA expression, processing and regulation in this disease. Our research is focused on 1) characterizing the molecular pathways and functions controlled by aberrant RNA splicing and 2) designing RNA- targeted therapies to uncover novel therapeutic vulnerabilities in this disease. Our group is heavily involved with RNA Sequencing of human and mouse pancreatic cancers. The candidate is expected to perform experiments exploring alterations in RNA splicing and additional steps in RNA processing, using a variety of preclinical model systems that include cell lines, xenografts, genetically engineered mouse models, and patient-derived models. A desire to develop and implement new methodologies is highly advantageous. There will be numerous opportunities for training to complement current skills, collaborate with other outstanding scientists, and to enhance grant writing skills. The candidate will work within an integrated, multidisciplinary team composed of basic scientists, computational biologists, pathologists, medical oncologists, and radiologists, with significant resources for bioinformatics support, access to clinical specimens, and studies in human and animal tissues. We have a long-standing interest in elucidating the molecular mechanisms of lung development, repair/regeneration, and tumorigenesis. These studies aim to establish the foundation for treating human diseases such as chronic obstructive lung disease (COPD) and lung cancer. To this end, we use the mouse as a model system and also take advantage of normal and diseased human lung tissues. We combine mouse genetics, cell biology, genomic analysis, CRISPR technology, proteomics, and lung organoids to address central questions in alveolar formation and repair, and the pathogenesis of lung cancer development. Our research program holds the promise of translating discovery from bench to bedside. The ideal candidate would have a strong expertise in bioinformatics and keen interest in themes of genomics, epigenetics, DNA methylation analysis, nuclear architecture, and aging. Our main research interests are (1) in situ structural study of important biological processes through cryo-electron tomography; and (2) development of correlative light and electron microscopy methods and further their applications. We are particularly looking for new members with good experience in either advanced light imaging, structural biology at near-atomic level, cell biology, physics, or material science. Experience in cryo-electron microscopy/tomography is not essential (we will teach you!), however, will be considered as an advantage. It takes cell models and mouse models of chromosome variation in cancer and genetic diseases as research objects to study the dose effect, timing regulation, organelle localization and protein-protein interaction of proteins in cancer. Introduction of Tutors and Laboratories Dr. Liu Yansheng He did postdoctoral research at the Federal Institute of Technology in Zurich, Switzerland, and joined the Institute of Cancer Biology of Yale University as an assistant professor at the end of 2017. Dr. Liu Yansheng, academic leader of the laboratory, specializes in quantitative proteomics DIA technology (Liu y. et al, cell2016, Liu y. et al, nature biotechnol, 2019) and applies it to phosphorylated proteome (rosenberger r r. *, Liu y. * et al. Nature Biotechnology 2017), plasma proteome (liu y.et al, molecular systems biology 2015) and down syndrome (liu y.et al, nature communications 2017), etc. In recent years, the first author or correspondent has published more than 30 articles in the magazines Cell, Nature Biotechnol, Nature Comm, PNAS, Molsystbio, Cell Rep, Molcell Proteomics, etc., with a total citation of more than 1600 times. He also served as editorial board of Proteomics, editorial board of Proteomics-Clinical Applications, and deputy editor-in-chief of Frontiers in Physiology.
