Position description
The Botchan Lab seeks candidates for appointment as postdoctoral scholar to join our project: Studies to Explore DNA Replication Proteins in Functional Assemblies through Intrinsically Disordered Domains. The Botchan Lab studies the study the mechanisms and regulation of the initiation of DNA replication in eukaryotes and how this process is coupled to the cell cycle. Drosophila melanogaster provides us with a unique metazoan model for this study as we have a genetic handle to readily test discoveries from biochemistry and structural biology. The primary objective of this project is the evaluation how intrinsically disordered regions are used to manifest phase-separated compartments or protein/nucleic- acid clustering for functional purposes. Initiator proteins ORC, CTD1 and Cdc6 all phase separate from bulk solutions in test tubes and are recruited to chromosomes in anaphase where such molecular attributes seem critical. In particular the ORC1 IDR is essential for DNA replication. ORC is also present in drosophila and human heterochromatin compartments with HP1 in G1 and S phases. How this interaction is mediated and for what ends will be a major goal.
